Course objectives
The course consisting of closed lectures introduces students with advanced analytical techniques and is upgrade to basic courses Analytical chemistry I a Analytical chemistry II. The new trends in analytical chemistry (development of instrumentation, methodology) are presented in lecture. In addition, the interdisciplinary character of present-day analytical chemistry is stressed. The lecture is documented by numerous examples taken from literature or practice.
Syllabus
  • 1. Kinetic methods of chemical analysis. Theory - knowledge of basic terms and their use in analytical chemistry. Instrumentation and their use for analysis.
  • 2. Examples of determination of inorganic and organic compounds. Examples of determination of biochemical compounds. Use of chemometric algorithms, computer applications.
  • 3. Chemical analysis in mixed and non-aqueous solutions. Titration in mixed and non-aqueous milieus, two-phase titration. Application of tenzides in analytical chemistry.
  • 4. Examples of optimisation of analytical methods (use of chemical modeling and chemometrics).
  • 5. Modern trends in chemical analysis (use of computers in "multi-array" data collection and their treatment, joining of analytical techniques - "hyphenated" techniques, miniaturisation, automation).
  • 6. Advanced optical methods and their use in analytical chemistry. Laser analytical spectroscopy - Properties and types of lasers. Analytical interaction of laser beam with matter. Linear methods - examples, optoacoustic, LIF spectroscopy, etc. Nonlinear methods. Special methods - detection of trace amounts, time-resolved spectroscopy, LIDAR far detection.
  • 7. Survey of instrumental methods suitable for organic analysis (Raman, IR, NMR spectroscopy, MS spectrometry, polarimetry, spectropolarimetry a CD spectrometry, refractometry a interferometry) and examples of use.
  • 8. Modern separation methods. Preconcentration of samples - Solid Phase Extraction (SPE). Membrane separation - ultrafiltration , dialysis, elektrodialysis, reverse osmosis. Methods with supercritical state of matter - extraction, chromatography. FFF methods.
  • 9. Analytical methods using radioisotopes. Basic terms. Methods. Description of methods and their use in practice.
  • 10. Surface analysis. Laser ablation. Techniques with photon probe (PES, XPS, UPS, LAMMS, etc.). Techniques with electron probe (EPXMA, SEM, kombinovaná AEM, AES, etc).
  • 11. Techniques with ion probe (RBS, ISS, SIMS, etc.). Techniques with pole probe (FIM, etc.). Microscopy with scanning probe (STM, AFM, etc.).
  • 12. Analytical X-ray spectrometry and their use in structural analysis.
