Completion of basic courses of molecular biology and plant cell biology is a prerequisite for subscription to the course. Completion of courses of plant physiology and plant anatomy will be an advantage.

Course objectives
The goal of the course is to understand and explain the nature of complex mechanisms and signalling pathways taking place within plant cells, and to correctly interpret their significance for physiological processes in plant tissues, organs and whole organism. Students will understand the advanced structures and physiological processes of plant cells.
Learning outcomes
At the end of the course, students will have an overview of: intracellular and intercellular signalling in plant cells, phytohormonal regulations, intracellular and intercellular transport, special mechanical properties of plant cells, plant cell cycle, plant cell differentiation in intact organism and in vitro, omics approaches on cellular level.
Syllabus
  • 1) Signalling pathways of phytohormones: auxin, cytokinin, ethylene, abscisic acid, gibberellins, brassinosteroids, jasmonates, salicylic acid and strigolactones; sensors
  • 2) Intracellular and intercellular transport: cell localization of metabolic enzymes, auxin transport, PIN proteins, PIN polarity and phosphorylation, cytokinin transporters, sugar transporters, plasmodesmata, symplast and apoplast, intercellular transport of nucleic acids
  • 3) Plant cell cycle: cell division as a response to environmental stimuli, plant cell cycle transitions, plant nuclear proteins, plastid division, mitochondrial and peroxisomal division, mechanism of organelle inheritance in dividing plant cells, cell division in nodulation, cell division affected by pathogens
  • 4) Plant cell enlargement: organization of the plant cell wall, biosynthesis and assembly of cellulose, signalling-dependent cytoskeletal dynamics and plant cell growth, the regulation of plant cell expansion-auxin-induced turgor-driven cell elongation, how phytohormones and their interactions affect cell growth
  • 5) Mechanical properties of plant cells: mechanosensors and signalling, gravity signal perception, amyloplasts, gravity signal transduction, gravity signal transmission, gravitropic growth response
  • 6) Plant stem cells: stem cell of shoot apical meristem, WUS-CLV3 signalling, Ca2+ function, stem cells of root apical meristem, WOX5-CLE, phytohormonal regulations, epigenetic regulations, de novo establishment of stem cells – lateral roots, phyllotaxis 7) Plant cell differentiation: asymmetric cell division in zygote, polarization, endoreduplication, procambium and cambium in plant vascular tissue development, programmed cell death, storage cells - oil and protein bodies
  • 8) In vitro plant cell differentiation: molecular aspects of dedifferentiation, redifferentiation, transdifferentiation, genetic reprogramming of plant cells in vitro, callus, regeneration, epigenetic modifications
  • 9) Plant single cell „omics“ I: transcriptomics, proteomics, metabolomics, lipidomics, synthetic biology, organellar omics studies
  • 10) Plant single cell „omics“ II: single cell and organellar hormonomics, cells sorting, nuclei sorting, cell organelle isolation, reporter cell lines, "cell atlas"
