Studium interakcí lidských telomerových proteinů

Abstract: Telomery jsou nukleoproteinové struktury, které se nacházejí na koncích lineárních chromozomů. Jejich hlavní funkcí je ochrana chromozomových konců před nukleolytickou degradací, opravnými mechanismy buňky a nechtěnou fúzí chromozomů. Kvůli neúplné replikaci při každém buněčném cyklu dochází ke zkracování telomer. Jejich opětovné prodlužování zajišťuje enzym telomeráza, který je schopen připojovat volné nukleotidy na konce chromozomů. Na regulaci telomerázové aktivity se významně podílejí proteiny shelterinového komplexu. Tyto proteiny mění prostorové uspořádání telomer tvorbou lasovitých struktur na jejich konci, čímž ovlivňují přístup životně důležitých enzymů k DNA a zprostředkovávají tak změnu signálních drah. Tímto způsobem shelterinové proteiny regulují zprostředkovaně délku telomer a podílejí se tak na procesech stárnutí a nádorové transformace buněk. Tato práce se zabývá studiem telomerových proteinů TRF2 a Rap1, jež jsou součástí shelterinového komplexu. V rámci práce byla určena afinita vazby proteinu TRF2 k DNA a vliv přítomnosti proteinu Rap1 na tuto afinitu. Kvantitativní analýzou pomocí anizotropie fluorescence a gelové retardační analýzy bylo zjištěno, že přítomnost proteinu Rap1 vede ke snížení vazebné afinity proteinu TRF2 k DNA a zároveň k významnému zvýšení selektivity proteinu TRF2 k telomerické DNA. …moreless

Abstract: Telomeres are nucleoprotein structures located at linear chromosome ends. The main fuction of telomeres is to protect chromosome ends from nucleolytic degradation, repair mechanisms of a cell and unwanted chromosome fusions. The telomeres shorten due to the incomplete DNA replication after each cycle of cell division. Telomere extension is provided by an enzyme called telomerase which is able to add free nucleotides to the ends of chromosomes. The regulation of telomerase activity is significantly influenced by proteins of the Shelterin complex. These proteins alter the spatial arrangement of telomeres by formation of lariat structures, thereby affect access of essential enzymes to DNA and mediate changes of signaling pathways. In this way, Shelterin proteins regulate telomere length and they participate in the processes of aging and tumor cell transformation. This work is focused on telomeric proteins TRF2 and Rap1 which are the components of Shelterin complex. The binding affinity of protein TRF2 to DNA and the effect of the presence of protein Rap1 on this affinity were determined in this work. The quantitative analysis using fluorescence anisotropy measurement and electrophoretic mobility shift assay revealed that the presence of Rap1 protein leads to a reduction in the binding affinity of protein TRF2 to DNA and also significantly increases the selectivity of protein TRF2 to telomeric DNA. …moreless
