Course objectives
The aim of this course is to explain principles of microscopy, methods of tissue processing and image analysis. Student will get the understanding about wide spectrum of microscopic techniques and their application. Student will learn methods of processing tissue samples – cells, tissues for individual microscopic techniques. Finally, student will be able to choose optimal method for detection of specific cell structures and molecules, able to explain and perform some of methods of processing of tissue samples for histological analysis with light microscopy.
Learning outcomes
Student will:
- understand wide spectrum of microscopic techniques and their application.
- know methods of processing tissue samples – cells, tissues for individual microscopic techniques.
- be able to choose optimal method for detection of specific cell structures and molecules
- be able to explain and perform some of methods of processing of tissue samples for histological analysis with light microscopy.
Syllabus
  • 1. Fundamentals of light microscopy – transmission light, dark field, contrast enhancement, stereomicroscopy, image acquisition
  • 2. Preparation of biological samples. Principles of histological staining.
  • 3. Fluorescence microscopy – epifluorescence, confocal, holographic, spectral microscopy, SPIM.
  • 4. Superresolution microscopy – SIM, STED, PALM, STORM.
  • 5. Live imaging of cells. Techniques. Staining of samples.
  • 6. Automated high content screening microscopy.
  • 7. Electron microscopy – principles, sample preparation, SEM, TEM, CryoTEM, SBF-SEM.
  • 8. Special microscopy – 2-photon, AFM, X-ray microscopy, microCT.
  • 9. Image processing and analysis.
