Course objectives
At the end of this practical course students should be able to:
describe the historical and present construction of light microscopes;
operate light microscopes in different modes and techniques;
prepare various kinds of botanical microscopic objects;
document these objects by means of microphotography;
process the microphotographs.
Syllabus
  • Introduction to the Methods for the Study of Plant Structure: Light Microscope Development, Structure and Function. Koehler Illumination.
  • Replicas: Study of the Surface Structure by Light Microscope. Stoma. Subsidiary and Neighbour Cells; Stoma Types.
  • Cleared Preparations: Angiosperm Embryo Development.
  • Squash Methods: Cell Cycle, Mitosis, Chromosome Staining.
  • Anatomical Methods: Fixation. Dehydration. Infiltration and Embedding. Paraffin Sections. Sections Staining and Mounting.
  • Vital Preparations: Phase Contrast Microscopy, Nomarski DIC.
  • Pollen: Differential Staining and Pollen Viability
  • Fluorescence Microscopy: Primary and Secondary Fluorescence, Immunofluorescence.
  • Documentation: Measurements. Photomicrography.
