Course objectives
At the end of this practical course students should be able to:
describe the development of the light microscope and actual construction of the light microscope;
operate the light microscopes in different modes and techniques;
use various methods for preparations from botanical objects;
document these objects by means of microphotography;
process the microphotographs.

  • Introduction to the Methods for the Study of Plant Structure: Light Microscope Development, Structure and Function. Koehler Illumination.
  • Replicas: Study of the Surface Structure by Light Microscope. Stoma. Subsidiary and Neighbour Cells; Stoma Types.
  • Cleared Preparations: Angiosperm Embryo Development.
  • Squash Methods: Cell Cycle, Mitosis, Chromosome Staining.
  • Anatomical Methods: Fixation. Dehydration. Infiltration and Embedding. Paraffin Sections. Sections Staining and Mounting.
  • Vital Preparations: Phase Contrast Microscopy, Nomarski DIC.
  • Pollen: Differential Staining and Pollen Viability
  • Fluorescence Microscopy: Primary and Secondary Fluorescence, Immunofluorescence.

Teaching methods
Presentations covering theoretical bases of different procedures for the study of plant tissue structures using the light microscope are used as an introduction of each lesson. Practical procedures of selected histological techniques lead to microscopic preparation which is analysed by means of proper method of the light microscopy. Plant structure is documented by drawing or as microphotography. Protocol of each procedure containing theoretical base as well as analysed results is prepared by each student as homework.

Control of protocols of used methods, written test, conclusion.
