Course objectives
Mutagenesis envolves basic lectures from molecular biology and genetics. The main goal of the discipline is to give to students a satisfactory deep uderstanding of the molecular mechanisms of spontaneous and induced mutagenesis. The subjects of this discipline also are the environmental genotoxicants, mutagenicity tests and strategies and methods for genetic risk assessment.
Syllabus
  • 1. History of mutagenesis, types of mutations. 2. Origin of spontaneous mutation. Tautomerization, deamination of DNA bases. Role of DNA repair, replication, and recombination. 3. Induced mutations. Radiation-induced mutations. Biological effects of UV radiation. Consequences of thinning of ozone layer. 4. Biological effects of ionizing radiation. Sources and types of ionizing radiation. Radiation-induced DNA damage. Factor controlling biological effects of radiation. Biological effects of magnetic field. 5. Chemical mutagenesis and chemical mutagens. Classification of chemical mutagens. 6. Promutagens. Metabolic activation of promutagens, plant promutagens, animal promutagens. Pharmacological polymorphisms. 7. DNA damage and repair mechanisms (fotoreactivation, excision repair, postreplication repair, SOS repair). Error prone and error free DNA repair, adaptive repair. 8. Molecular basis of mutagenesis induced by physical and chemical mutagens. 9. Antimutagens. Classification of antimutagenic compound. Mechanisms of antimutagenenesis. 10. Environmental mutagens and carcinogens. 11. Method for genetic risk assessment. Identification of mutagens among potentially hazardous chemicals. 12. Monitoring environmental genotoxicant. 13. Principles of mutation tests.
