Course objectives
Basics of analytical separation methods, transit from liquid-liquid extraction to chromatography. Efficiency of separation methods. Sample preparation for analytical separation. Theory and praxis of Liquid Chromatography (LC). Ion exchange chromatography (IEC) and other variants of LC (SFC, HILIC, etc). Chiral separations. Separation of macromolecules (GPC, HDC, FFF).
Syllabus
  • 1. introduction; extraction: description of extraction equilibria in system liquid-liquid 2. description of extraction equilibria in system liquid-solid 3. preseparation; preanalytical sample preparation; SFE (supercritical fluid extraction); ASE (accelerated solvent extraction); MAE (microwave assisted extraction) 4. TLC (thin-layer chromatography); SPE (solid phase extraction); HSE (head-space extraction) 5. analytical separation; chromatography theoretical ground; classification of basic separation mechanisms in liquid chromatography LC (liquid-liquid chromatography, LLC, liquid-solid chromatography, LSC, ion chromatography, IC, gel permeation chromatography, GPC) 6. thermodynamics of separation; kinetics of separation 7. LC instrumentation; mobile phase delivery; injection devices; separation column, detectors (diode array detectors, fluorescence, amperometric, conductivity, refractometric) 8. LC methods: analytical information in chromatogram; RP-HPLC (reverse phase liquid chromatography), IEC (ion exchange chromatography), 9. chromatofocusation, affinity chromatography; SFC (supercritical fluid chromatography); PC (perfusion chromatography) 10. chiral separation: definition of chirality; optical rotation dispersity; chiral selectors and their aplication in separation methods (CE, HPLC) - (Dr. Pazourek) 11. separation of macromolecules: radius of gyration; relative molecular mass, polydispersity; optical detection, viscosimetry; ultracentrifugation, gel capillary electrophoresis; matrix assisted laser desorption ionization-time of flight (MALDI-TOF) - (Dr. Pazourek) 12. gel permeation chromatography (GPC), hydrodynamic chromatography (HDC), field-flow fractionation (FFF); special detection techniques; static light scattering (LS), low-angle laser light scattering (LALLS), multi-angle laser light scattering (MALLS); hyphenation of separation and detection techniques. - (Dr. Pazourek)
