Course objectives
The main aim of the course is to present current developments in the field of liquid-based chromatography. The special attention will be paid to individual retention mechanisms, new types of stationary phases, elevated temperature, gradient elution, and multidimensional separations. Part of the course will be focused on miniaturization and unusual detection.
At the end, student will be able understand current liquid chromatography.
Learning outcomes
After the course student will:
- know the most comon separation mechanisms;
- know how to characterize HPLC column;
- know new types of stationary phases;
- apply knowledge about gradient elution, high temperature separations and multidimensional techniques;
- provide orientation in miniturization of separation techniques;
- be able to compare several detection methods applicable in HPLC;
- be able to optimize isocratic and graient elution;
Syllabus
  • 1. Retention mechanisms – principle of chromatography, instrumetnation, sumnmary of basic mechanisms (NP, RP, HILIC, IEX, IC, SEC)
  • 2. Column characterization – porosity, permeability, efficiency, resolution, kinetic plot
  • 3. Silicagel – structrue, modification, fully porous, nonporous, superficially porous particles
  • 4. Inorganic monoliths – preparation, modification, application
  • 5. Polymer monolilths – classification, preparation, modification, application
  • 6. Gradient elution - models, prediction, peak compression, application
  • 7. Elevated temperature - entropy, enthalpy, quality of separation, thermal gradients
  • 8. Ultra high pressure - analysis time, instrumentation, application
  • 9. Multidimensional chromatography – theory, optimization, sample transfer, application
  • 10. Miniaturization - nano LC, chips, integration, uTAS
  • 11. (Unusual) detection – UV, MS, (elektrochemistry, NMR, IR, …)

Teaching methods
The course is presented as weekly presentations focused on individual parts of advanced liquid chrmatography.
At the end of each class, designated time for discussion will be allowed.
Assessment methods
The course is finished with both written and oral exam.
At first, short test with 10 questions is answered. Following oral examination with two questions is allowed only in more than five questions are answered correctly.
