The target audience is BSc and MSc students. During the course, all necessary information will be provided. However, a basic knowledge of genetics is welcome.

Cíle předmětu
This course will use various laboratory techniques to test the segregation law of genetics as established by G. Mendel, the correlation between genotype and phenotype, and the effects of plant hormones for plant development. The lectures are accompanied by practical work. In case of eLearning and distance-teaching imposed by the COVID situation, only the theorical part will be taught remotely via Teams.
Výstupy z učení
Students should be able to understand the analysis of segregation for a single genetic character, and the link between genotype and phenotype. Students will learn basic lab techniques. Students will be initiated to the role of plant hormones in plant development.
Osnova
  • Using Arabidopsis mutants the students will observe and calculate the frequencies of the mutant’s phenotypes, and correlate it with the genotype of individual seedlings. This part of the practical uses laboratory techniques such as work in sterile conditions, sow seeds for in vitro culture, monitoring seedling growth and analysis of the seedling phenotypes by stereomicroscope.
  • In a second part of the course, the student will investigate the effects of the plant hormone auxin on root growth (few concentrations tested) and on reporter lines for the hormone signalling pathway. Activation or repression of the signalling pathways will be tested by colorimetric staining. Results will be collected and conclusion will be written and discussed. This part of the practical includes tissue culture as in the first part (including preparation of the growth medium), result analysis (root growth measurements), colorimetric staining and microscopy.
