Course objectives
This course is intended for Bc., MSc. and PhD. students and would give an overview of recent molecular techniques used in entomological research. At the end of the course, students should be able to understand the theoretical and practical principles of following subjects: DNA analysis (extraction of DNA, PCR, electrophoresis, sequencing, fragment analysis of microsatellites), Analysing methods (phylogenetic data: distance methods, maximum parsimony, model testing, maximum likelihood, bayesian inference, etc.).
Syllabus
  • Lectures 1. DNA (history, chem. structure, types of DNA, character of primers and their testing) 2. Methods based on nucleic acid analyses (PCR, MS, RAPD, AFLP, hybridization) 3. DNA sequencing 4. Barcoding and Tree of Life – use of molec. data, multigene phylogeny, working with databases 5.Rules of the work safety in a molec. biology laboratory Practice- LAB 6. Lab work I (DNA extraction from insect tissue) 7. Lab work II ( PCR, gel electrophoresis) 8. Lab work III (sample preparation for sequencing, purification and sequencing) 9. Lab work IV (mikrosatellites and fragment analysis) Practice - PC 10. Analysis methods I (sequnecing data correction) - analyses based on sequnece fragments of chosen gene markers 11. Analysis methods II (phylogenetic data) - methods based on distnace, parsimony, likelihood, bayesian inference, model testing

Teaching methods
The course will provide classic lectures as well as practical lab work with own material and computer based analyses with obtained data.
Assessment methods
Students will be classified according to their work skills, results in the laboratory and in data analyses. For the successfull ending of the course, a 100% presence is required.
