Course objectives
Students will acquire a working knowledge of high throughput, high resolution techniques of protein separation and identification in complex biological samples. The techniques form a base for a global analysis of a protein complement of expressed genome - proteome. Further, the students will master basic techniques of protein structural biology, including production in bacterial cells of eukaryotic proteins as fusion proteins, and their subsequent single-step affinity chromatography purification, and structure-function characterization.
Syllabus
  • Introduction to proteome analysis. Methods of proteome analysis. Electrophoretic techniques - 1-D, 2-D PAGE, IEF. Mass spectrometry techniques - MALDI-TOF, ESI-MS. Proteome databases. Recombinant protein technology. DNA sequence analysis. Chromatography techniques. Effects of protein fusions. Image analysis.
