Course objectives
Separation methods play significant role not only in analytical chemistry but also in all other chemistry branches, in chemical and biochemical technology and pharmaceutical industry. Capillary electrophoresis is modern separation method and one of the most efficient separation techniques, generally. The main advantage besides its high efficiency is low consumption of the sample and possibility to separate even very similar compounds and various isomers including the chiral ones. The main goal of the course is to explain the students the basic principles in such way that they would be able on the base of gained knowledge to use rationally the technique in real practice and to know how to apply it even to the separation problems not described in the literature: for separation and determination of inorganic, organic, pharmaceutics and drugs and also to use it in other branches like biology, medicine and technology. Therefore, part of the course is also practical exercise and problem solving on a computer and simulation programs afforded with extensive database of compounds, theirs physico-chemical constants, etc. which allows to learn the students to think. The course is also accompanied with basics of the electrophoretic methods development and optimization including the use of a combination of the experimental design and with the use of artificial neural networks.

  • History of development. Basics of electromigration Placement of Capillary electrophoresis in separation methods Basic modes of electrophoresis Basic terms: ionic mobility, effective mobility Calculation of ionic and effective mobility, determination Electroosmotic flow (EOF), principles. Stern double layer; Zeta potential Possibilities how to measure EOF Modification of EOF with various approaches Principles of the instrumentation, scheme of the instrument, capillary dimensions, detection windows Technical details; uncoated and coated capillaries Detectors and detection techniques in CE Uv Vis, DAD, potentiometric, conductivity, mass spectrometry and laser induced fluorescence Hyphenation of CE and MS ev. CE and MALDI Methods of sample injection in CE; hydrodynamic and electrokinetics; differences. Basic electrolyte; pH changes, various buffers Dispersion, main factors Ohm law, effect of temperature, elimination-cooling Electrokinetic micellar chromatography, principals and separation of neutral compounds Chemical equilibria and kinetics in CE Effect of chemical reactions on the separation;the role of complexation and its use Chiral separation, chiral selectors, possibilities of the influence to chiral separation Various applications of CE ; brief survey Optimization in CE; basic possibilities; Single-factor experiments; multivariate approaches and artificial neural networks Possibilities of next development; chip electrophoresis.
