Course objectives
The lecture course is focused on acquiring theoretical knowledge needed for implementation of modern spectroscopic methods for analysis of protein-protein and protein-DNA interactions. From the methodical approaches, the main emphasis is put on highly sensitive fluorescent methods and their practical use in biological laboratories. The main lecture topics are detection, techniques for fluorescent labeling of macromolecules and their application for analysis of function and interaction of important biomolecules in genome and proteome.

  • Steady-state fluorescence
  • Time-resolved fluorescence
  • Fluorescence anisotropy and its change after interaction of macromolecules
  • Absorption spectroscopy in biological analysis of molecules
  • Fluorescence quenching, resonance energy transfer - application for studies of structural changes of biomolecules
  • Intrinsic fluorescence of biologically important molecules
  • Extrinsic fluorescence – fluorescent labels, probes, and indicators
  • Fluorescence labeling of DNA and proteins
  • Fluorescence microscopy
  • Analytical applications of fluorescence for concentration determination
