Abstract
Haloperidol is an older antipsychotic used in the treatment of schizophrenia and in the treatment of acute psychotic states and delirium. Due to its strong central antidopaminergic action, it is classified as a highly potent neuroleptic. However, the usage of typical neuroleptic drugs has been limited by the side effects and toxicity produced by them. Therefore is essential to know all possible risks which bring together treatment with these drugs. We decided to measure and compare level of oxidative stress from plasma and erythrocytes from 17 guinea pigs (10 treated and 7 untreated). We were concerned on superoxide dismutase detection,FRAP method, glutathione reductase and reduced and oxidized glutathione GSH:GSSG ratio measurement as a relevant oxidative stress pointer.We treated 17 guinea pigs by haloperidol and measured oxidative stress from plasma and isolated erythrocytes. Haloperidol was present in plasma of treated animals while was undetectable in animals without treatment. In animals treated by haloperidol was significantly increased superoxide dismutase (SOD) and glutathione reductase, which is clear evidence about ROS and oxidative stress. SOD was increased also in isolated erythrocytes and also ratio between reduced and oxidized glutathione(GSH:GSSG) was shifted to oxidative stress state. The present study demonstrates that administration of haloperidol causes significant oxidative stress.
