Abstract |
---|
We have identified a new putative lectin from Pisum sativum using SDS-PAGE and MS analysis of highly purified pea amine oxidase where it was present as an impurity. This study was aimed at the production of this protein in a eucaryotic expression system and its structure-function characterization. As production of the native protein was found very low, we have compared two yeast expression systems - Pichia pastoris and Saccharomyces cerevisiae. Advantages/disadvantages of both approaches will be discussed in detail. |

Protein predikovaný jako lektin byl nalezen pomocí SDS-PAGE a MS analýzy při purifikaci aminooxidázy z hrachu, kde byl přítomen jako nečistota. Protože produkce nativního proteinu v prokaryotním organismu Escheriachia coli byla velmi nízká, byly vybrány dva systémy kvasinkových expresních organismů - Pichia pastoris a Saccharomyces cerevisiae. Studie je zaměřena na porovnání exprese cílového proteinu ve dvou eukaryotnich expresních systémech, další práce bude pokračovat funkčními studiemi a určení struktury proteinu. |
