Abstract
The contribution of immunoecological studies has gained a place of central importance in recent evolutionary biology and ecology. The main aim of the work is to analyze the associations between multiple parasites and host immunocompetence and to investigate the structure and evolutionary mechanism of selection in MHC genes of cyprinid fish species. The extremely high level of allelic diversity in the genes of major histocompatibility complex results of many mechanisms and is determined with the intent to obtain genetic data for evolutionary and population studies. Nowadays, for MHC genotyping we use two basic approaches: 454 clonal pyrosequencing of amplicons and CE-SSCP of amplicon clones combined with automatic sequencing based on Sanger technology. In this contribution, we present primary results of a new approach for MHC genes variability tests – high resolution melting analysis (HRM) and expression studies (using qPCR), that enable us to extend the studies on MHC genes up to functional level.
