Abstract
Thanks to exceptional affinity of plant alkaloids to various biomolecules they can be used in analytical chemistry as a probes. Many isochinoline alkaloids are fluorescent and interact with DNA by intercalation. Phenantridine alkaloids such as sanguinarine, chelerytrine, chelirubine, chelilutine, sanguilutine and macarpine interact with double stranded DNA showing substantial changes in fluorescence emission. The response of luminescence intensity to low concentrations of double stranded DNA in presence of alkaloids was found to be linear and can be potentially used for quantification of DNA by means of luminescence spectrometry. For example, limit of detection (3σ method) calculated for alkaloid chelirubine was calculated to be 8 ng.mL -1 of DNA. Chelirubine and macarpine were even successfully used for fluorescent DNA-specific staining of living cells due to the lower toxicity. In conclusion, we have demonstrated that natural products, phenantridine alkaloids, can be used as alternative probes to synthetic DNA dyes such as ethidium bromide.
