Abstract
Domnelá hydroláza TmpE bakteriofága 812K1-420 sa modifikovala histidínovou kotvou a po expresii prečistila metódou metalo-afinitnej chromatografie a gélovej filtrácie. Výsledný produkt vykazuje dostatočnú čistotu pre analýzu metódami kryštalografie. Zistenie presnej štruktúry pomôže detailne popísať bakteriofága 812 na atomárnej úrovni. Prečistený produkt sa taktiež využíva pre funkčnú analýzu TmpE, stanovovaním jeho aktivity voči kmeňu S. aureus 1137, či už priamo na miskách alebo nepriamo voči stafylokokovým bunkovým stenám na zymograme (nepublikované výsledky). Prípadné potvrdenie lýtickej aktivity môže viesť k použitiu tohto proteínu ako enzybiotika.
Abstract (in English)
This study is part of a research which is dealing with reconstruction of whole virion structure of bacteriophage 812, attacking wide spectrum of clinical and commensal strains of pathogen Staphylococcus aureus, including those which are resistant to antibiotics. The concrete aim consists in detail characterization of putative hydrolase TmpE, which is part of the phage tail. The hydrolases due to their efficacy in disposal of bacteria cell walls have been applied in number of industrial and clinical fields as enzybiotics, the proteins killing pathogen agents. Thus, detail analysis could prove predicted function of TmpE protein and lead to development of protein-based product efficient against S.aureus.
