Abstract
In this chapter, we describe a site-directed mutagenesis method used for investigating the sugar binding pattern of the PA-IIL lectin from the pathogenic bacterium Pseudomonas aeruginosa . Moreover, procedures for the production and purification of PA-IIL mutants are described, and several basic methods for characterizing the mutants are discussed.

V této kapitole popisujeme metodu místně-cílené mutageneze, která byla použita pro studium vazebné specifity lektinu PA-IIL, který pochází z bakterie Pseudomonas aeruginosa. Nadto, jsou zde popsány procedury pro produkci a purifikaci mutantů lektinu PA-IIL a je zde diskutováno několik základních metod pro charakterizaci těchto mutantů.
