The SMC complexes (cohesin, condensin and SMC5/6 complex) are highly conserved molecular machineries and the key components of higher order chromatin structures. The cohesin complex mediates sister chromatid cohesion from S-phase until anaphase, whilst condensin promotes chromatin condensation during mitosis. The SMC5/6 complex is essential for proliferation in yeast and is involved in the non-essential homologous recombination-based DNA repair processes, including repair of DNA double strand breaks, restart of stalled replication forks etc. However the precise mechanisms of its involvement in these biological processes remain unclear. To uncover its essential function, we decided to do yeast S.pombe synthetic lethal genetic screen with hypomorphic nse3/R254E DNA-binding mutation, which reduces association of the SMC5/6 complex with chromatin. Detailed genetic analysis revealed that HAT module of the SAGA H3 histone acetyltransferase complex negatively interacts with nse3/R254E and smc6 hypomorphic mutations. Using Y2H analysis, we found physical interactions between HAT module and SMC5/6 subunits. Genetic data suggest that the essential function of the SMC5/6 complex is likely connected with HAT module of the SAGA chromatin modifying complex.
