Abstract
Phosphorylation of STAT3 at Serine 727 and its nuclear translocation indicates direct and indirect induction of regeneration program in the rat dorsal root ganglia neurons Kokošová V., Klusáková I., Hradilová-Svíženská I., Dubový P. Department of Anatomy, Faculty of Medicine, Masaryk University, Brno, Czech Republic Phosphorylation of STAT3 on serine 727 (pSTAT3-S727) can modulate transcriptional activity of STAT3 phosphorylated on position of Tyrosine 705 (pSTAT3-Y705). Evidence of pSTAT3-S727 expression was not yet investigated in the dorsal root ganglia (DRG) neurons. Sixteen rats of Wistar line (male, 250-300g) were divided into four groups. The right sciatic nerve of the first group was compressed by triple ligatures (SNC, n=4), the sciatic nerve of the second group was cut and ligated to avoid spontaneous reinnervation (CSNT, n=4), and the sciatic nerve of the third group rats was only exposed without any damage (sham-operation). All rats were left to survive for 7 days. Rats without any surgical intervention were used as a naive control. Animals were perfused by Zamboni fixation and DRG were removed bilaterally in segments L4 and C6-C7. Expression of pSTAT3-S727 was detected by indirect immunohistochemical method and analyzed by image analysis program (NIS Elements, Nikon). Low intensity of pSTAT3-S727 immunofluorescence (IF) was measured in the neuronal nuclei of the lumbar and cervical DRG of the naive rats. Significant increased intensity of IF was found bilaterally in neuronal nuclei of DRG at L4 as well as C6-C7 segments after SNC and CSNT when compared with DRG from naive or sham-operated animals. The present results of pSTAT3-S727 expression in DRG neurons correlate well with our published results of IL-6, IL-6R and pSTAT3-Y705 elevation in both lumbar and cervical segments after unilateral sciatic nerve injury and support the idea of spreading inflammatory signaling alongside the spinal cord.
