Abstract

V originále

This work deals with the evaluation of lipophilicity and stability of selected carbamates using high performance liquid chromatography. A method using C18 reverse stationary phase and isocratic conditions was used to determine lipophilicity. The methanol:water mobile phase was used to determine the capacity factor k, while the methanol:acetate buffer (pH 6.5 and pH 7.4) was used to determine the distribution coefficient D. Prediction of lipophilicity by in silico methods was performed using three software with different algorithms: ChemBioDraw Ultra, ACD/Percepta and MOLinspiration. The carbamate with the weakest steric protection, i.e. with the expected highest degradability, was chosen for stability monitoring. The developed HPLC method using phenyl stationary phase and acetonitrile:phosphate buffer (pH 2.5) as the mobile phase provided sufficient resolution between the studied carbamate and its degradant, pattern anilide.
