In rheumatoid arthritis (RA) and bronchial asthma (BA), uPAR plays an important role, however the impact of uPAR splicing variatns on the disease development and severity has not been investigated yet. We analysed the PLAUR splicing pattern changes in RA and BA patients´ monocytes and macrophages with both mild and severe clinical phenotype and compare with healthy individuals. The results to this date indicates potential changes in variant frequency encoding full form of suPAR in monocytes of RA patients compared to monocytes isolated from BA patients and healthy volunteers. Another change in the splicing pattern is observed for the variant frequency encoding the ΔE5+6 membrane-bound uPAR in monocytes of BA patients compared to monocytes isolated from RA patients and healthy volunteers. In all tested groups, the decreasing character in alternatively spliced isoforms proportion during monocyte-to-macrophage differentiation can be seen. Due to important role of both macrophages and monocytes in RA and BA, the expression profile of uPAR could have an impact on disease course an/or it could be an important marker of their inflammatory status.
