Influence of metal ions on activity and stability of DNase I immobilized on hydrophilic particles

The aim of work has been concentrated on degradation of high-weight and plasmid DNAs using immobilized DNase I. The activity of free and immobilized DNase I was estimated from UV spectrophotometric measurements and from agarose gel electrophoresis. The immobilized enzyme was full actived using Co2+ ions as an activator, a decrease of the activity was observed using other ions. Practical applications of DNase I immobilized on poly(HEMA-co-EDMA) microspheres in cleavage of plasmid DNAs are discussed.
