Abstract
The genus Armillaria belongs to basidiomycetes and has been known to induce root rot disease and to cause extensive economic losses to a forest crop. More than 40 isolates containing six species of Armillaria from the collections have been analyzed (A. borealis, A. cepistipes, A. gallica, A. mellea, A. ostoyae and A. tabescens). We have used the internal transcribed spacer (ITS), which is highly polymorphic and provides useful tool for taxonomic studies thanks to highly conserved sequences of these region. The resolution achieved on the HPLC system was comparable or better than that achieved by electrophoresis and HPLC has better performance in quantitation. Moreover, the restriction analysis of the isolates using HPLC allowed to clearly differentiate some heterozygots from homozygots.
