Abstract
Experimental design combined with artificial neural networks enables faster optimization of the separation conditions than classical single variable approach. HPLC can be successfully used for the determination of the state of 2-deoxycytidine 5-monophosphate methylation in DNA. Utilization of ED-ANN within HPLC has greatly enhanced the method of quantitative determination of 5-methyl-2'-deoxycytidine 5'-monophosphate and 2'-deoxycytidine 5'-monophosphate in the cell homogenate. Use of this approach has allowed us to optimize separation of several types of mixtures containing purine and pyrimidine bases, their nucleosides and nucleotides.

Plánování pokusů kombinované s umělými neuronovými sítěmi umožňuje rychleji optimalizovat separační podmínky než klasický přístup změnou jedné proměnné. HPLC může být použita pro stanovení stavu metylace 2-deoxycytidin-5-monofosfátu v DNA. Využitím ED-ANN pro HPLC se zvýšila citlivost stanovení 5-metyl-2'-deoxycytidin-5'-monofosfáte a 2'-deoxycytidin-5'-monofosfátu v buněčném homogenátu. Použití tohoto postupu jsme mohli optimalizovat separci několika druhů směsí purinových a pyrimidinových bazí, jejich nukleosidů a nukleotidů.
