Abstract
A series of twenty biotechnologically usable Lactobacillus strains with unclear taxonomical position were analysed and characterized by biochemical tests as well as by ribotyping. All strains were characterized by using API 50CH kit and by conventional tests key for this bacterial group. Biochemical test results assigned 17 strains to the species L. acidophilus; two strains were identified as L. rhamnosus and one strain were identified only as Lactobacillus sp. Ribotyping with EcoRI and a probe complementary to 16S and 23S rRNA divided all strains analysed into a few groups. Both L. rhamnosus strains as well as one Lactobacillus sp. strain were differentiated in the full agreement with biochemical test results. Ribotyping revealed intraspecies diversity among seventeen L. acidophilus strains and divided this group into four clusters. The most numerous cluster including eleven strains and the type strain L. acidophilus CCM 4833T. Obtained results suggested that ribotyping as applied in this study could be useful for differentiation of L. acidophilus complex. This study was supported by Ministry of Agriculture project QE 1382.

Biotechnologicky využitelné kmeny rodu Lactobacillus s nejasnou taxonomickou pozicí byly identifikovány biochemickými testy a ribotypizací. Všechny kmeny byly charakterizovány soupravou API 50CH a konvenčními testy. Tyto výsledky zařadily 17 kmenů jako L. acidophilus; dva kmeny jako L. rhamnosus a dva kmeny byly identifikovány pouze jako Lactobacillus sp. Ribotypizace s EcoRI a sondou komplementární k 16S a 23S rRNA rozdělila identifikované kmeny do několika poodskupin. Oba kmeny L. rhamnosus i dva kmeny určené pouze jako Lactobacillus sp. byly odlišeny v plné shodě s výsledky biotypizace. Ribotypizace ukázala rovněž vnitrodruhovou variabilitu mezi 17 kmeny L. acidophilus, které rozdělila do čtyř skupin. Nejpočetnější skupina zahrnovala 11 kmenů včetně typového kmene L. acidophilus CCM 4833T. Získané výsledky naznačují že ribotypizace je užitečná metoda pro diferenciaci kmenů komplexu L. acidophilus.
