Abstract
his study deals with the separation of basic polycyclic aromatic nitrogen heterocycles (PANH) by reversed-phase high-performance liquid chromatography (RP-HPLC) on two octadecylsilane stationary phases made by different manufacturers. The aims of our investigation were to evaluate surface properties of the selected sorbents (hydrophobicity and silanol activity) and to compare the retention behavior of tested PANH on these stationary phases. Experiments were carried out with the following members of basic PANH: quinoline and its mono-methyl derivatives, isoquinoline, benzo[h]quinoline, acridine and phenanthridine. Zorbax Eclipse XDB C18 (Agilent Technologies) and Purospher Star RP-18e (Merck) both in 250 x 4,6 mm package with 5 um i.d. were used as octadecylsilane stationary phases. The evaluation procedures as well as the separation experiments studying retention behavior of basic PANH were carried out under isocratic elution conditions. Evaluation of both reversed-phase sorbents was performed via empirical method based on chromatographic data of the test compounds chosen according to the hydrophobicity and silanol activity of tested sorbents surfaces. During separation experiments several different elution conditions were applied on each column. Especially buffer concentration its pH and ionic strength in conjunction with pKa values of individual heterocycles were taken into consideration.
