Abstract
The use of free lanthanide ions and their complexes for plasmid DNA pBR322 and chromosomal DNA was studied. Plasmid pBR322 DNA was treated by lanthanide chlorides (Eu3+, La3+, Nd3+, Pr3+, Gd3+ ) in HEPES buffer (pH 7.0, 7.5 and 8.0) at different temperatures. The formation of linear and nicked plasmid forms was investigated depending on the reaction conditions. Heterogeneous lanthanide complexes of ethylenediamine tetraacetic acid (EDTA) immobilized on insoluble methacrylate support and iminodiacetic acid (IDA) immobilized on styrene support were used as catalysts plasmid for DNA pBR322 cleavage, too. The temperature of reaction mixture had substantial influence on cleavage rate. The precipitation of DNA occured during the measurement of interactions between chromosomal DNA and La3+ ions.

Bylo studováno použití volných lanthanidových iontů a jejich komplexů ke štěpení plazmidové pBR322 a chromosomální DNA. Plazmidová pBR322 DNA byla štěpena při různých teplotách chloridy lanthanidů (Eu3+, La3+, Nd3+, Pr3+, Gd3+ ) v HEPES pufru (pH 7,0; 7,5 and 8,0). Byla studována tvorba plazmidových forem (linearizované a s jednořetězcovým zlomem)v závislosti na použitých reakčních podmínkách. Jako katalyzátory při štěpení plazmidové pBR322 DNA byly rovněž použity heterogenní lanthanidové komplexy ethylendiamino tetraoctové kyseliny (EDTA),imobilizované na nerozpustném methakrylátovém nosiči a iminodioctová kyselina (IDA), imobilizovaná na styrénovém nosiči.
