The neurons shift from normal maintenance and neurotransmission toward survival and regeneration after axonal injury. However, scarce information is available on reaction of satellite cells following axotomy. The present experiments demonstrate changes of immunofluorescence staining for laminin-1, chondroitin sulfate proteoglycan (CSP), tenascin (TEN), thrombospondin (THS) and fibronectin (FN) in the DRG after intercostal nerve ligation. Twenty adult rats were deeply anesthetized and left intercostal nerves (8-12) were ligated by 6-0 Mersilk thread. Animals were allowed to survive for 1 or 2 weeks. The corresponding DRG (Th8-12) of both sides were removed and immersed in Zamboni fixation. Intact DRG (Th8-12) were removed and fixed from other 4 rats. Longitudinal cryostat sections (12 ľm) through ipsilateral, contralateral and intact DRG were incubated for indirect immunofluorescence staining to detect laminin-1, CSP, TEN, THS and FN. An intensity of immunofluorescence staining was compared between digitalized pictures of ipsilateral and contralateral DRG by image analysis system (Lucia G). Ligature of the intercostal nerve induced enlargement of satellite cell sheaths in the ipsilateral DRG. In comparison to the contralateral and intact DRG, a laminin immunofluorescence was reduced around the neuron-satellite cell units in some regions of the sections through ipsilateral DRG. In contrast, immunofluorescence brightness for CSP, TEN, THS and FN was increased in enlarged satellite cell sheaths of ipsilateral comparing contralateral DRG. The quantitative changes of ECM molecules may be involved in external conditions that stimulate a growth of sympathetic sprouts in DRG after compression of peripheral nerve. ER -
