Abstract
The mechanism of ajmaline-induced inhibition of the transient outward current Ito has been investigated in right ventricular myocytes of rat using the whole cell patch clamp technique. Ajmaline decreased the amplitude and the time integral of Ito in a concentration-dependent, but frequency- and use-independent manner. We conclude that in the rat right ventricular myocytes ajmaline is an open channel blocker of Ito with fast recovery from the block at resting voltage.
Abstract (in Czech)
V této práci jsme se zabývali studiem vlivu ajmalinu na přechodný draslíkový proud z buňky Ito u pravokomorových srdečních buněk potkana metodou vnuceného napětí na celé buňce. Ajmalin snižoval amplitudu a časový integrál Ito v závislosti na aplikované koncentraci látky, avšak nezávisle na frekvenci stimulace či na předchozí stimulaci. Můžeme uzavřít, že u buněk z pravé komory potkana ajmalin blokuje Ito přes otevřený stav kanálu s rychlým zotavením z blokády při klidovém napětí.
