Lignans play important roles in plant defense based on their profound biological activities, which include antioxidant, antifungal, antibacterial, antiviral, insect antifeedant, phytotoxic and cytotoxic properties (Mi Kwon 2001, Gang 1999). These biological effects are closely related with hepatoprotective and anticancer activities of medical plant Schisandra chinensis (Chen et al. 1997, Opletal et al. 2001). While mainly present in heartwood and bark tissues, lignans can also accumulate in other organs and tissues including sapwood, leaves, fruits, seeds and roots. In vitro plant cultures can be potential sources of these unique natural substances. In this study the production of main dibenzo[a,c]cyclooctadiene lignans in dicotyledonous plant Schisandra chinensis was studied. S. chinensis calluses, derived from endosperm and immature zygotic embryo were developed on MS (Murashige, Skoog, 1962) and VW5 (Coke, 1996) medium supplemented with growth regulators (thidiazuron, 2,4-dichlorophenoxyacetic acid and benzyl aminopurine). High performance liquid chromatography on reversed phase was applied for profiling of lignans in various stages of the culture. Kubo et al. (1992) reported increased level of polyamine putrescine in liver under Schisandra lignans treatment. On the other hand putrescine in our experiments used as a supplement of the cultivation medium in concentration from 300 to 500 ěM increased the level of lignans in tissues. The brownish tissues showed their highest amount, particularly of deoxyschizandrin and gomisin N. Using of plant cell culture for the production of lignans seems to be one of the methods capable to improve yields of individual lignans. Kranz and Petersen (2003) successfully used in vitro suspension culture of Linum with relatively large brownish aggregates for lignan production. The determination of lignans by improved HPLC method is susceptible for selection of the best embryogenic lines of Schisandra chinensis and their further breeding.
