Abstract
Staphylococcus xylosus and Staphylococcus equorum are two biochemically and phylogenetically close species. Athough S.xylosus is usually only transient on human and is primarily acquired from domestic animal and their products, it has been isolated also from human clinical specimens, mainly from urinary tract infection. Lack of the biochemical key tests makes differentiating of these species difficult. It is very important to find another reliable approach to distinguish S.xylosus and S.equorum species. A group of 18 presumptive S.xylosus strains from human clinical isolates were analysed by biochemical tests and ribotyping. SDS-PAGE protein analysis was performed to confirm the results of ribotyping. Biochemical properties were tested by API Staph and ID 32 STAPH commercial kits as well as by conventional tests. Ribotyping was done with EcoRI and HindIII restriction enzymes and a probe complementary to 16S-23S rRNA. Electrophoresis of proteins was performed using a 5% polyacrylamide stacking gel and 12% separated gel containing 0.1% SDS. Biochemical tests identified 17 S.xylosus strains and one intermediated S.xylosus/S.equorum strain. Ribotyping in contrast to biotyping recognised 4 strains of S.equorum and one strain that shows high similarity to Staphylococcus nepalensis reference strains. Protein analysis divided tested strains into four groups. A group with four S.equorum strains, a second group which probably contains one S.nepalensis strain. Our data indicate that SDS-PAGE as well as ribotyping have got a higher discriminative power than biotyping to distinguish S.xylosus and S.equorum strains and that are able to identified them.

Skupina 18 presumptivních kmenů Staphylococcus xylosus pocházejících z humánního klinického materiálu byla charakterizováno za použití biotypizace, ribotypizace a analýzy celobuněčných proteinů metodou SDS-PAGE. Analýzou proteinů a také ribotypizací byly kmeny identifikovány jako zástupci druhu S.xylosus, 4 kmeny náležely do taxonu S. equorum a jeden kmen vykazuje podobnost k S. nepalensis referenčním kmenům.
