Abstract
In our work, we present a general, indirect method, using which it is possible to quantify the percentage of the given amino acid modified in total in a protein sequence (protein modification degree, PMD). From the knowledge of the identity of amino acid residues modifiable during post-translation process, PMD and its changes during time of particular amino acid residue can be determined. Such a quantification can be achieved by means of easy stable isotope modification (via 18O-labelling by trypsination) and consequential MALDI TOF MS analysis.

Tato práce prezentuje obecnou, nepřímou metodu, pomocí níž je možné stanovit rozsah celkové modifikace dané aminokyseliny proteinu (protein modification degree, PMD). Známe-li identitu aminokyseliny modifikovatelné posttranslační modifikací, PMD a její změny ve vztahu k těmto aminokyselin mohou být takto stanoveny. Toto stanovení je dosaženo snadným značením stabilními izotopy (značení 18O v průběhu stěpení trypsinem) a následnou MALDI TOF MS analýzou.
