Abstract
We have isolated symbiotic bacteria from the entomopathogenic nematode Heterorhabditis bacteriophora H221 (Pouzdřany). Isolate Photorhabdus sp. CCM 7002 shows clear luminescence which is characteristic for genus Photorhabdus, the only known terrestrial luminescent bacteria. Thus, our isolate was compared with type cultures of Photorhabdus species (P. asymbiotica ssp. asymbiotica, P. asymbiotica ssp. australis, P. luminescens ssp. akhurstii, P. luminescens ssp. kayaii, P. luminescens ssp. laumondii, P. luminescens ssp. luminescens, P. luminescens ssp. thracensis, P. temperata). Phenotypic characteristics were based on identification kits (API 50 CH, API 20 NE, ENTEROtest 24) supplemented with additional conventional tests. The most closely related or similar species to Photorhabdus sp. CCM 7002 based on 98 phenotypic characteristics appears to be P. luminescens. To compare the protein spectrum of studied bacterial species, bacterial proteins were analysed by acrylamide gradient gel electrophoresis (SDS-PAGGE). We found more than 40 protein fractions in the range of 6.5 - 200 kDa. Our data demonstrated significant differences among protein profiles of Photorhabdus taxa. Finally, the 16S rRNA gene sequence comparison of isolate Photorhabdus sp. CCM 7002 and Photorhabdus type strains exhibited 100 % identity with P. luminescens ssp. kayaii, which was described on the basis of two isolates from Turkey, 2004. The strain Photorhabdus sp. CCM 7002 is the first known isolate of this subspecies from the Czech Republic and probably also the second record described globally. This study was supported by grant MSM0021622416.

Izolovali jsme symbiotické bakterie z entomopatogenní hlístovky druhu Heterorhabditis bacteriophora H221 (Pouzdřany). Izolát Photorhabdus sp. CCM 7002 vykazoval jasnou bioluminiscenci, která je charakteristická pro rod Photorhabdus. Proto byl náš izolát srovnáván s typovými kulturami všech druhů rodu Photorhabdus (P. asymbiotica ssp. asymbiotica, P. asymbiotica ssp. australis, P. luminescens ssp. akhurstii, P. luminescens ssp. kayaii, P. luminescens ssp. laumondii, P. luminescens ssp. luminescens, P. luminescens ssp. thracensis, P. temperata). Fenotypová charakteristika byla založena na identifikačních kitech(API 50 CH, API 20 NE, ENTEROtest 24) společně s konvenčními identifikačními testy. Nejpodobější našemu izolátu Photorhabdus sp. CCM 7002 byl na základě 98 charakteristik P. luminescens. Proteinové spektrum bakterií bylo determinováno akrylamidovou gradientovou elektroforézou (SDS-PAGGE). Nalezli jsme více než 40 proteinových frakcí v rozmezí 6.5 - 200 kDa. Na základě této metody jsou jasně odlišitelné jednotlivé druhy rodu Photorhabdus. Nakonec použitím 16S rRNA genové sekvence byla prokázána 100 % shoda s P. luminescens ssp. kayaii, který byl popsán na základě dvou izolátů z Turecka v roce 2004. Náš izolát Photorhabdus sp. CCM 7002 je prvním izolátem tohoto poddruhu v České republice možná také druhým izolátem celosvětově. This study was supported by grant MSM0021622416.
