Abstract
The genetic analysis of a newly identified accession of wild barley (Hordeum vulgare ssp. spontaneum) resistant to powdery mildew (Blumeria graminis f. sp. hordei) showed that the resistance was determined by two dominant genes. The aim of the molecular analysis was to localize the resistance genes on barley genetic map using DNA markers (SSRs mostly). The analyses were performed in the F2 population of the cross between the winter barley cultivar Tiffany and the resistant accession PI466197. The susceptible F2 plants were screened to identify the markers linked with the resistance genes. Data from F2 plants of all genotypes were analysed in the MapManager QTX program to confirm the linkage and to assign the order and the distances between the markers. The molecular analysis revealed a highly significant linkage with Bmac0213 and MGB402 on the chromosome 1H, which is the position consistent with the Mla locus. The other gene was proved to be linked with Bmac0134 and MWG878 on the short arm of chromosome 2H. The prospect of the work is to find more tightly linked DNA markers so that breeders could use them for marker assisted selection.

U nově identifikovaného zdroje rezistence PI466197 (Hordeum vulgare ssp. spontaneum) k padlí travnímu (Blumeria graminis f. sp. hordei) byla genetickou analýzou stanovena přítomnost dvou dominantních genů rezistence, z nichž jeden se nachází v lokusu Mla. Molekulární analýzou pomocí DNA markerů byla určena pozice jednoho genu na krátkém rameni chromozomu 1H mezi markery Bmac0213 a MGB402 (oblast lokusu Mla) a druhého genu na krátkém rameni chromozomu 2H mezi markery Bmac0134 a MWG878.
