Homologous recombination (HR) plays a key role in the elimination of cytotoxic and genotoxic lesions induced by ionizing radiation, carcinogenic chemicals, and chemotherapeutic compounds. Furthermore, cells have to rely on homologous recombination to rescue stalled replication forks and hence prevent fork demise. If not processed properly, DSBs can lead to chromosomal aberrations and rearrangements, which then give rise to strong mutator and cancer phenotypes. Genetic analyses in the yeast (Saccharomyces cerevisiae) have revealed a central role of the Srs2 helicase in preventing untimely HR events and the formation of potentially deleterious DNA structures upon DNA replication stress. In contrast, Srs2 function appears to be important also for proper repair of DNA double-strand breaks by HR. In this work, we aim to dissect the multi-functional role of Srs2 in HR and other cellular processes using combination of biochemical, genetic and structural approaches.
