Abstract
Association of single nucleotide polymorphisms (SNPs) T-786C and G894T in gene encoding eNOS with blood pressure variability (BPV) in man was evaluated. Systolic and diastolic blood pressure variability in relative and absolute units were determined in 129 healthy subjects by spectral method as spectral power at the frequency of 0.1 Hz. Genotypes were detected by means of polymerase chain reaction and restriction analysis. Carriers of less frequent alleles showed significantly greater BPV comparing more frequent alleles homozygotes. The determined SNPs were in linkage disequilibrium. Carriers of genotype CC (for T-786C) with TT (for G894T) displayed significantly greater BPV compared to carriers of other genotype combinations. We found significant associations of T-786C and G894T polymorphisms in gene encoding eNOS with systolic and diastolic blood pressure variability. Less frequent alleles of T-786C and G894T in gene encoding eNOS are associated with increased systolic and diastolic BPV.
Abstract (in Czech)
Cílem studie bylo vyhodnotit asociaci polymorfismů T-786C a G894T v genu pro eNOS s variabilitou v krevním tlaku (VTK) u člověka. Relativní a absolutní variabilita v systolickém a diastolickém tlaku krve byla určena u 129 zdravých osob spektrální metodou při frekvenci 0,1 Hz. Genotyp byl stanoven metodou polymerázové řetězové reakce a restrikční analýzy. Nositelé méně četných alel měli signifikantně větší VTK ve srovnání s homozygoty s více četnými alelami. Studované polymorfismy byly ve vazebné nerovnováze. Největších nositelé Nositelé genotypové kombinace CC (pro T-786C) s TT (pro G894T) dosahovali signifikantně vyšších hodnot VTK než nositelé ostatních genotypových kombinací. Byla nalezena signifikantní asociace polymorfismů T-786C a G894T v genu pro eNOS s variabilitou v systolickém a diastolickém krevním tlaku.
