We demonstrated different immunostaining for CB2 receptor in the DRG of adult rats by two antibodies raised against different epitopes of CB2. Rats were operated for unilateral sciatic nerve ligation and allowed to survive for 1 week. Longitudinal cryostat sections through ipsi-and contralateral L4 and L5 DRG were incubated with different primary antibodies. Immunofluorescence for CB2 receptors was observed predominantly in the nuclei of the DRG neurons when a rabbit polyclonal antibody raised against CB2 of human origin was used as the primary antibody. Using a goat polyclonal antibody raised against CB2 of mouse origin displayed immunostaining in the cytoplasm of the DRG neurons whereas nuclei were not stained. Total immunoreactivity of both antibodies was increased in the neurons of both ipsi- and contralateral DRG comparing with intact ones. ER -
