Abstract
Magnetic nonporous P(HEMA-co-EDMA) particles were prepared by dispersion copolymerization of 2-hydroxyethyl methacrylate (HEMA) and ethylene dimethacrylate (EDMA) in the presence of maghemite nanoparticles. Magnetic nonporous P(HEMA-co-GMA) microspheres were prepared by dispersion copolymerisation of 2-hydroxyethyl methacrylate (HEMA) and glycidyl methacrylate (GMA) in the presence of colloidal oleic acid-coated magnetite (Fe3O4) particles. Subsequently, the hydroxy groups of the particles were oxidized with 2 wt.% aqueous solution of potassium permanganate under acidic conditions (2 M sulphuric acid). Prepared particles were used for isolation of bacterial DNA from real samples. DNA was adsorbed on particles in the presence of 8 wt. % of poly(ethylene glycol) (PEG 6,000) and 2 M NaCl concentrations. DNA was eluted using TE buffer and used directly for PCR amplification and identification Lactobacillus and Bifidobacterium cells in dairy products, Trichophyton cells in lyophilisates, bacterial cells in sediments from Lake Baikal.

Magnetické neporézní částice P(HEMA-co-EDMA)disperzní kopolymerizací 2-hydroxyethyl methacrylátu (HEMA) a ethylen dimethacrylátu (EDMA)v přítomnosti nanočástic maghemitu. Magnetické neporézní mikročástice disperzní kopolymerizací 2-hydroxyethyl methacrylátu (HEMA) a glycidyl methakrylátu (GMA) v přítomnosti koloidních částic megnetitu smočeného kyselinou olejovou.Hydroxylové skupiny částic byly oxidovány 2% vodným roztokem manganistanu draselného v kyselém prostředí (2 M kyselina sírová) Takto připravené částice byly použity k izolaci bakteriální DNA z reálných vzorků. DNA byla adsorbována na částice v přítomnosti 8% poly)ethylen glykolu) (PEG 6000) a 2 M NaCl. Dna byla eluována pomocí TE pufru a přímo použita v PCR. Byly identifikovány buňky bakterií rodu Lactobacillus a Bifidobacterium v mléčných výrobcích, buňky houby Trichophyton v lyifilizátech a bakteriální buňky v sedimentech z jezera Bajkal.
