Abstract |

Amphibian populations are declining worldwide, evidently as the implication of environmental contaminants and infectious diseases. Generally, oxidative stress is considered to be one of the major toxicity mechanisms, however, only few amphibian studies are dealing with detoxification responses besides standard parameters. The aim of this study was to estimate the influence of widespread environmental pollutant in standard acute test using Xenopus frogs supplemented with the measurement of biomarkers of oxidative stress. Sublethal effects on modulation of antioxidative and detoxification compounds (glutathione, glutathione-S-transferase, glutathione reductase and peroxidase, catalase, superoxide dismutase and cytochrome P450 as EROD) were observed. Oxidative stress expressed as the potential lipid and protein dissociation was determined. Lower effect concentrations of biomarkers of oxidative stress preceding any sign of toxicity were found. The role of oxidative stress in embryotoxicity and teratogenesis was examined. Research was supported by the chancellor of Masaryk University (project 20061431E0014) and INCHEMBIOL 0021622412. |

V posledních letech se diskutuje o možném negativním vlivu pesticidů v souvislosti se zmenšováním populací a četnějším výskytu malformací volně žijících obojživelníků. Ačkoliv je v tomto procesu zapojeno více environmentálních faktorů, některé studie a fakta poukazují na škodlivé působení pesticidů na raná stádia obojživelníků obecně. Negativnímu vlivu na vývoj obojživelníků nahrává i shoda období páření obojživelníků s dobou aplikace postřiků na zemědělské plochy, kde dochází k depozici do půdy a následnému transportu do povrchových vod, které tvoří přirozený habitat juvenilních stádií obojživelníků a také prokazatelná citlivost těchto raných stádií vůči pesticidům. Cílem této studie bylo zhodnotit toxicitu paraquatu v testu FETAX, kde bylo kromě hodnocení klasických embryotoxických parametrů zahrnuto biochemické stanovení antioxidativního a detoxifikačního systému s cílem posoudit, zda publikovaná embryotoxicita herbicidu paraquatu je spojena s indukcí oxidativního stresu. Byl posuzován projektivní účinek externího přídavku známého antioxidantu, vitamínu C, ve směsi s paraquatem na ovlivnění hladiny oxidativních markerů. |
