Abstract
S. chinensis cultures derived from immature zygotic embryo were developed on Murashige and Skoog and VW5 medium. Embryogenic culture was established on the medium containing thidiazuron, 2,4dichlorophenoxyacetic acid and benzylaminopurine. An analytical method based on HPLC was used for the determination of six lignans in plant cell cultures. We have explored the possibility of increasing the lignan release to medium by addition of two neutral polymeric resins (Amberlite XAD2 and XAD7) to the cell cultures. The polymeric resins not only notably enlarged portions of extracellular lignans, but also significantly enhanced production of total lignans. Amberlite XAD2 increased production of deoxyschizandrin, whereas Amberlite XAD7 enhanced the amount of wuweizisu C. Lignans adsorbed on polymeric resins can be easily recovered and purified without destruction of the cells. Lignans were also tested for their cytotoxicity on HeLa cells, A 431 human squamous cell epithelioma and human skin fibroblast. The nonpolar lignan gomisin N was found to be more cytotoxic than other lignans tested.
