Abstract

Pochopení hematoonkologických onemocnění na proteinové úrovni je významné pro předpověď citlivosti na podanou terapii, možnost diagnostiky onemocnění či vývoj cílené léčby a musí být podloženo znalostí molekulární patogenese nádoru. Proteomika popisuje analýzu kompletního proteomu buňky nebo tkáně a zahrnuje v sobě mnoho technologií_ Western blotting, dvourozměrnou elektroforézu v polyakrylamidovém gelu, hmotnostní spektrometrii, protein_ čipové technologie. Ačkoli dosud bylo provedeno jen několik proteomických studií u hematoonkologických onemocnění, potenciál těchto technologií umožňuje objevení nových biomarkerů spojených s rezistencí na podané léčivo a identifikaci biomarkerů, které mohou usnadnit vývoj rychlých diagnostických testů, snadno aplikovatelných v klinickém nastavení. Tento článek proto popisuje používané proteomické techniky a upozorňuje na práce věnované proteomickému výzkumu hematoonkologických onemocnění, zejména publikací věnovaných výzkumu mnohočetnému myelomu, leukémií a lymfomů.

Abstract (in English)

The understanding haematological malignancies at the protein level is important for the prediction of therapy response, the possibility for disease diagnostics, and the development of targeted treatment and must be based on knowledge regarding the molecular pathogenesis of the tumour. "Proteomics" describes the analysis of the entire proteome of a cell or a tissue and incorporates multiple technologies, including - Western blotting, two-dimensional polyacrylamide gel electrophoresis, mass spectrometry, and ProteinChip-based technology. Although there is a limited number of studies to date in haematological malignancies, those performed highlight the potential impact of these technologies in the discovery of novel markers associated with the drug resistance and the identification of biomarkers which may facilitate the development of a rapid diagnostic test easily applicable in the clinical setting. Thereby this article describes used proteomics techniques and highlight studies regarding proteomic research in haematological malignancies, primarily publications concerning research of multiple myeloma, leukemias and lymphomas.
