TY - JOUR ID - 726038 AU - Španová, Alena - Rittich, Bohuslav - Horák, Daniel - Rybnikář, Alois PY - 2007 TI - Isolation of PCR-ready DNA from real samples containing PCR inhibitors PB - Research Institute of Chemical and Process Enginnering, University of Pannonia, Vesazprém, Hungary CY - Budapest, Hungary SN - 9789639696235 KW - fungi KW - Trichophyton KW - nonporous magnetic hydrophilic microspheres KW - P(HEMA-co-EDMA) KW - P(HEMA-co-GMA) N2 - Hydrophilic magnetic particles coated by carboxyl groups were used for PCR-ready DNA isolation. DNA was adsorbed on magnetic particles in the presence of 8 wt. % of poly(ethylene glycol) (PEG 6,000) and 2 M NaCl concentrations, washed and eluted into TE buffer (10 mM Tris-HCl, pH 7.8; 1 mM EDTA; pH 8.0). The eluted DNA was used directly for PCR amplification and identification of Lactobacillus and Bifidobacterium cells in dairy products (cheese, fermented dairy products) and lyophilisates, Trichophyton cells in mixed cultures and bacterial cells in sediments from Lake Baikal. It was also used in GMO testing in foods with success. ER -
