Abstract

V originále

RecA test is a short-term luminescent SOS genotoxicity test. The induction of the SOS system is detected through the activation of the reporter gene system - DNA damage SOS repair system. The test is based on the swift elevation of recA gene expression in response to DNA damage. In case of RecA test there is a specific recA::luxCDABE gene fusion. It is fusion of reporter gene and specific gene of SOS repair system. This gene fusion could detect genotoxicity efficiently. In this study Escherichia coli strains DPD2794, DPD2797, DPD1718 and DPD1710 were used for detection of genotoxicity. The genotoxicity was detected by monitoring of bioluminescent emission. For assessment of the sensitivity evaluation a group of chemicals and environmental samples were selected. The aim of this research was to decide whether this test has future applications in the screening evaluation of genotoxic potential and to compare sensitivity of the RecA test with various strains of Escherichia coli. Results indicate that the strain DPD2797 could be recommended for testing of environmental samples and also pure chemical substances, because this strain showed most sensitive responses to selected chemical standards and also to environmental samples.
