MALDI MS of intact bacterial cells or their lysates appears to be a highly promising tool for bacterial identification. During the past decade, numerous laboratories demonstrated the ability of this technique to rapidly resolve bacterial samples, concerning mostly a model set of organisms characterised previously or in parallel by other taxonomical approaches. Almost every laboratory developed its own sample preparation protocol working in a satisfactory way on a particular system of interest. Many factors were found to influence the signal quality, sensitivity, reproducibility and other method characteristics resulting in notable changes in MALDI MS profiling discriminatory power.
