Abstract
The interaction of BCL-2 G-quadruplex forming sequence with cationic porphyrin TMPyP4 and its C-rich complementary strand was studied by means of molecular absorption spectroscopy and circular dichroism, experimental dates were analysed by advanced chemometric techniques based on mulitvariate analysis. Analyses resulted in proving 1:2 stoichiometry for the complex BCL-2G:TMPyP4 and equilibrium constant 5,0*10^13 M-2, that is weaker than complex of BCL-2 complementary strands, equilibrium constant of which was determined 10^7,7 M-1.

Studie interakcí oligonukleotidu bohatého na guanin, tvořícího G-kvadruplex, s kationickým porfyrinem TMPyP4 a komplementárním vláknem DNA. Experimenty byly monitorovány pomocí molek. absorpční spektroskopie a cirkulárního dichroismu, analýza dat pokročilými chemometrickými metodani založenými na multivariátní analýze. Výsledky potvrdily navrženou stechiometrii komplexu BCL-2G:TMPyP4 1:2, s konstantou stability 5,0*10^13 M-2. Duplex obou komplementárních sekvencí BCL-2 je silnější, konstanta stability 10^7,7 M-1.
