Abstract |

Many environmental (pesticides, fertilizers) and anthropogenic (trematode infections, the loss of habitat, UV) factors are possibly involved in the global amphibian decline. Some pesticides (e.g. bipyridyls) are designed to produce ROS and initiate the process of oxidative stress also in nontarget species such as amphibian. This oxidative damage of macromolecules can possibly lead to disturbance of normal developmental processes, moreover when the amphibian breeding period is parallel to pesticide application onto agricultural land. The aim of this study was to estimate the influence of two widespread bipyridyls: paraquat and diquat on the early developmental phases of Xenopus laevis in FETAX test with emphasis on their oxidative action. Besides the standard FETAX parameters the sublethal effects on modulation of antioxidative and detoxification compounds (glutathione, glutathioneStransferase, glutathione reductase and peroxidase) together with the level of lipid peroxides were studied. The protective role of the external addition of antioxidant ascorbic acid was examined. Significant decreases in embryotoxicity and teratogenity linked with modulation of biochemical parameters after ascorbic acid application have been observed confirming the oxidative mechanism of action of used pesticides. |

Existuje mnoho environmentálních (pesticidy, hnojiva) a antropogenních (infekce, ztráta habitatu, UV záření) faktorů potenciálně zodpovědných za celosvětový pokles populace obojživelníků. Některé pesticidy (napřílad bipyridyly) jsou designovány k produkci volných kyslíkových radikálů, které iniciují proces oxidativního stresu, avšak také v necílových organismech, jako jsou obojživelníci. Toto oxidativní poškození makromolekul může teoreticky vést až k narušení normálních vývojových procesů, a to zvlášť v případě shody aplikace postřiků na polní kultury s obdobím páření obojživelníků. Cílem této studie je posoudit vliv dvou široce používaných bipyridylů paraquatu a diquatu na časné vývojové procesy drápatky Xenopus laevis v testu FETAX s důrazem na jejich oxidativní působení. Vedle standardních parametrů byly studovány subletální efekty; modulace antioxidativních a detoxifikačních látek (glutation, glutationStrasferáza, glutation reduktáza a peroxidáza) společně s úrovní lipidní peroxidace. Také byl pozorován účinek externího přídavku antioxidantu vitamínu C, po kterém se významně snížila embryotoxicita a teratogenita společně se změnou biochemických parametrů, což potvrzuje oxidativní mechanismus účinku testovaných pesticidů. |
