Abstract
In cultivations of the most common experimental plant A. thaliana, chemical control is unevoidable. We invesigated co-effects of fungicides/insecticides and high light/heat treatments. Wild type A. thaliana (Columbia) and LHC-defficient mutants (Lhcb 2-1) were used and treated each 30 d by the following chemicals: (1) Previcur 607SL, (2) Zolone 35EC, (3) a mixture of 0,25% Previcur and 0,2% Zolone, and (4) untreated control. Additionally, the plants were exposed to 2 short-term treatmets: (a) 1000 micromol m-2 s-1 of PAR (high light = HL) for 60 min and (b) combination of HL and 30 oC. Treatment effects on photosynthesis were monitored by a PAM-2000 fluorometer. Before and after the treatments, pigments were extracted from leaf tissue and HPLC determined: contents of chla, chlb, viola-, anthera-, zeaxanthin, lutein, beta-carotene were evaluated by a HPLC (Waters). Results showed that Previcur plus Zolone caused oxidative stress in chloroplasts even in control plants indicaded by increased values of zeaxanthin and DEPS. In Lhcb mutants, Fv/Fm decreased after HL and temperature treatments which was attributed to negative short-term effects in photosystem II. Supported by FRVS 1546/2008.
